StreptaClick® HRP Multiplex IHC Kit is an accelerated heat-treatment-free multiplex Tyramide Signal Amplification (TSA) staining solution with imaging for up to six colors for both frozen and FFPE samples in one day. This comprehensive kit is designed for researchers who demand ease of use, flexibility, precision and reliability in immunohistochemistry (IHC).

The kit includes everything needed to label biotinylated antibodies with horseradish peroxidase (HRP) enzyme in solution utilizing our modified streptavidin StreptaClick®. As StreptaClick® removes the issue of antibody aggregation through monovalent biotin binding it is particularly suited for labeling primary biotinylated antibodies. The kit also features a novel HRP block buffer that eliminates the need for antibody-stripping by heat-treatment such as boiling between cycles. This feature maintains tissue morphology over multiple staining cycles making it ideal for both frozen and FFPE tissue sections, and higher degrees of multiplexing than comparable solutions.

In combination, these features makes the protocol automation friendly and results for six color + DAPI experiments are achievable in one day.

How it works

Biotinylated antibodies pre-mixed with StreptaClick® operate similarly to directly conjugated antibodies, thereby eliminating the need for secondary antibodies. The StreptaClick® labeling reaction is extremely efficient, permitting the labeling of any antibody volume, even down to microliters. This is particularly beneficial when setting up new experiments.

Quick Results, Less Hands On Time

With StreptaClick® HRP you can complete a 7-color Tyramide Signal Amplification (TSA) multiplex IHC within a single day (DAPI + 6 biotinylated antibodies). Each biotinylated antibody is labeled with HRP by the fast StreptaClick® reaction, and the kit includes an HRP block buffer that efficiently quenches HRP activity between staining cycles. This avoids the slow, inconvenient and harsh boiling step to strip off antibodies required by other solutions between staining cycles. Added benefits of the HRP block buffer include preserved tissue morphology, higher attainable protein biomarker multiplexing levels and a protocol that is automation-friendly.

Seven-Color TSA method for frozen sections from mice, humans, rats, and other species, and FFPE

The StreptaClick® HRP solution is a must for any manual or automated TSA staining. The kit comes with a HRP quenching buffer that eliminates the need for boiling, which not only accelerates each color cycle but also makes the kit highly beneficial for frozen tissue sections. The quenching buffer effectively silences the HRP signal from the previous round while preserving the morphology of the frozen tissue. Hence, the StreptaClick® HRP kit provides a rapid method for multiplex TSA IHC on both FFPE and frozen sections, with antibodies from any host, and on tissues from mice, humans, rats, and other species.

Rapid manual or automated protocol

Automation is also facilitated by the rapid protocol and unique features. The use of primary antibodies and our HRP quenching buffer makes it especially amenable for automated staining. External partners have developed a protocol on the Leica Biosystems BOND RX IHC staining system, and protocols for other systems are in development.

Is StreptaClick® HRP heat-free TSA suitable for beginners?

Yes, StreptaClick® HRP heat-free TSA is easy to use and well-suited for those new to the field. The heat-free protocol considerably reduces the optimization time required to build a multiplex antibody panel.

How does StreptaClick® HRP heat-free TSA address workflow and reproducibility challenges?

Through its TSA and HRP block system, the need for secondary antibodies is eliminated. This improves reproducibility and streamlines the workflow compared to indirect immunofluorescence (IF), while preserving target epitopes and tissue morphology.

How long does an experiment take?

Up to 5-7 antibodies can be applied within a single day depending on the required incubation time of each antibody, significantly reducing the total experiment time. On average one staining cycle takes 1.5 hours.

Does StreptaClick® HRP heat-free TSA require biotinylated antibodies?

Yes, StreptaClick® HRP heat-free TSA has been developed and validated for use with biotinylated primary antibodies. Biotinylated primary antibodies can be purchased from a number of vendors including our listed partners, either off the shelf or as custom orders. Alternatively, carrier free antibodies (ie no carrier protein such as BSA, and ideally free from sodium azide) can easily be biotinylated with our BiotinPure™ Antibody Biotinylation Kit. BiotinPure™ is an easy and complete solution only requiring a benchtop centrifuge.

StreptaClick® HRP Multiplex IHC kits

The kits contain StreptaClick® HRP antibody labeling reagent for labeling of biotinylated antibodies, and a novel horseradish peroxidase (HRP) block buffer. The staining protocol does not require heat treatment such as boiling between cycles, and is ideal for direct labeling of primary antibodies for higher degrees of multiplexing compared to working with secondary antibodies.

  • Tyramide 3-color Dye kit: includes Tyramide 488, Tyramide 555, Tyramide 647 and Tyramide Amplification Buffer Plus.
  • Tyramide 5-color Dye kit: includes Tyramide CF®430, Tyramide 488, Tyramide 555, Tyramide 594, Tyramide 647, and Tyramide Amplification Buffer Plus.
