The StreptaClick® 8-Plex Multiplex IHC Kit provides a rapid and heat-treatment-free Tyramide Signal Amplification (TSA) staining cyclic workflow for frozen and FFPE tissue sections. It is optimized for imaging on the ZEISS Axioscan 7 spatial biology platform, enabling visualization of up to eight colors in a single run. The kit is ideal for researchers in high-throughput laboratories who require a reliable, time-efficient, and flexible multiplex immunohistochemistry workflow.

Next-Generation TSA Staining – Powered by StreptaClick® HRP

 

The workflow is based on the StreptaClick® technology, which uses a modified streptavidin to link reporter molecules to primary biotinylated antibodies in solution.

The StreptaClick® HRP workflow transforms TSA staining into a heat-free, faster, and simpler process for both manual and automated applications, offering four key advantages over conventional protocols:

  1. Significantly shortened TSA workflow. The StreptaClick® technology eliminates the use of secondary antibodies, accelerating the protocol while maintaining full flexibility in primary antibody species selection.
  2. A heat-free workflow that preserves epitopes and tissue morphology. The proprietary HRP blocking buffer is specifically optimized to ensure efficient HRP inactivation while maintaining optimal tissue integrity – even on frozen tissue sections.
  3. Improved resistance to tyrosine depletion. The heat-free workflow does not strip off antibodies. As a result, new tyrosine residues are incorporated into the tissue with each antibody, making the method less susceptible to tyrosine depletion.
  4. Easy multiplex panel setup. By reducing epitope exhaustion and tyrosine depletion, the staining conditions remain more consistent across cycles. Consequently, the order of antibodies does not need to be optimized, which saves considerable time when establishing new panels.

StreptaClick® 8-plex Workflow

Labeling your primary antibodies with StreptaClick® HRP

Biotinylated antibodies pre-mixed with StreptaClick® HRP function in the same way as directly conjugated antibodies, eliminating the need for secondary antibodies. The StreptaClick® labeling reaction is highly efficient, allowing you to label any antibody volume — even as little as a few microliters. This makes it especially useful for testing new experimental setups or labs often changing antibody panels for different projects.

Multiplex immunofluorescence protocol

  1. Apply the first antibody to the tissue
  2. Develop color with TSA reagents
  3. Quench HRP activity with HRP block buffer
  4. Repeat the cycle with the next antibody until all have been developed

Each cycle takes approximately 1 hour to complete. Tyramide Signal Amplification with our carefully chosen tyramide dyes and amplification buffer provides 5-20X signal amplification over directly labeled antibodies.

Automatically scan the slide in the ZEISS Axioscan 7 spatial biology once all tyramide dyes have been developed.

Fast results, less hands on time

With StreptaClick® 8-plex, you can complete a 7-color Tyramide Signal Amplification (TSA) multiplex IHC assay in a single day (DAPI + seven biotinylated antibodies). Each biotinylated antibody is rapidly labeled with HRP through the StreptaClick® HRP reaction. The kit also includes a specialized HRP block buffer that effectively quenches HRP activity between staining cycles, eliminating the need for the slow, harsh boiling step typically required to strip antibodies between cycles. One cycle thereby only takes 1 hour, compared to 2.5 hours for traditional TSA workflows.

This HRP block buffer not only simplifies the workflow but also preserves tissue morphology, enables higher levels of protein biomarker multiplexing, and ensures compatibility with automated systems.

The streamlined protocol and innovative features of the StreptaClick® 8-plex kit make it well-suited for both manual and automated staining. By using primary antibodies along with the included HRP quenching buffer, automation becomes straightforward and efficient.
