StreptaClick® Precision is developed for site-specific conjugation with a defined number of attachment groups for click chemistry – currently exactly one azide for the possibility of achieving a 1:1 ratio of reporter to binder. Furthermore, StreptaClick® Precision has a unique cis-divalent design with maximum separation of the site for conjugation and biotin binding. This design makes StreptaClick® Precision ideal for attachment of bulky molecules without affecting the biotin binding affinity and without causing aggregation when mixed with biotinylated proteins as it still exhibits monovalent binding properties.
StreptaClick® Precision is currently provided with a single azide group for either Copper(I)-catalyzed azide-alkyne cycloaddition (CuAAC) or copper-free Strain-promoted azide-alkyne cycloaddition (SPAAC) click chemistry. We observe the highest efficiencies when utilizing SPAAC to react azide to dibenzylcyclooctyne (DBCO), especially when clicking oligonucleotides to StreptaClick® Precision.
Off-the shelf StreptaClick® Precision constructs are also available. These are pre-conjugated with a single 14 mer DNA oligonucleotide or a single bright fluorochrome (488 nm or 647 nm).
StreptaClick® Precision with a DBCO group for copper free click chemistry as well as pre-conjugated flurorescent proteins (phycoerythrin), enzymes (ex. horseradish peroxidase (HRP)) are in development. To achieve greater reporter to binder ratios we are also developing versions with exactly three and seven reactive groups.
External partners have successfully attached other molecules, including lipid nanoparticles (LNPs).
