StreptaClick® HRP Multiplex IHC Kit is an accelerated heat-treatment-free multiplex Tyramide Signal Amplification (TSA) staining solution for up to nine markers (8 antibodies + DAPI) for fixed cells, FFPE samples and frozen tissues. This comprehensive kit is designed for researchers who demand ease of use and flexibility in multiplex immunohistochemistry (IHC).
The kit includes everything needed to label biotinylated primary antibodies with horseradish peroxidase (HRP) enzyme in solution utilizing our modified streptavidin StreptaClick®. As StreptaClick® removes the issue of antibody aggregation through monovalent biotin binding it is particularly suited for labeling primary biotinylated antibodies. The kit also features a novel HRP Quench Buffer that eliminates the need for antibody-stripping by heat-treatment such as boiling between cycles. This feature maintains tissue morphology over multiple staining cycles making it ideal for both frozen and FFPE tissue sections, and higher degrees of multiplexing than comparable solutions.
In combination, these features makes the protocol highly automation-friendly, while results for manual staining of six markers + DAPI are achievable in one day.

How it works
Biotinylated antibodies pre-mixed with StreptaClick® operate similarly to directly conjugated antibodies, thereby eliminating the need for secondary antibodies. The StreptaClick® labeling reaction is extremely efficient, permitting the labeling of any antibody volume, even down to microliters. This is particularly beneficial when setting up new experiments.
Quick Results, Less Hands On Time
With StreptaClick® HRP you can complete a 7-color Tyramide Signal Amplification (TSA) multiplex IHC within a single day (DAPI + 6 biotinylated antibodies). Each biotinylated antibody is labeled with HRP by the fast StreptaClick® reaction, and the kit includes an HRP Quench Buffer buffer that efficiently quenches HRP activity between staining cycles.
Quench HRP activity avoids the slow, inconvenient and harsh boiling step to strip off antibodies required by other solutions between staining cycles. Added benefits of the HRP Quench Buffer include preserved tissue morphology, higher attainable protein biomarker multiplexing levels and a protocol that is automation-friendly. This also results in that optimization of the order in which antibodies are added to the tissue is virtually eliminated.
Nine-Color TSA method for frozen sections and FFPE from mice, humans, rats, and other species
The StreptaClick® HRP solution is a must for any manual or automated TSA staining. The kit comes with a HRP quenching buffer that eliminates the need for boiling, which not only accelerates each color cycle but also makes the kit highly beneficial for frozen tissue sections. The quenching buffer effectively silences the HRP signal from the previous round while preserving the morphology of the frozen tissue. Hence, the StreptaClick® HRP kit provides a rapid method for multiplex TSA IHC on both FFPE and frozen sections, with antibodies from any host, and on tissues from mice, humans, rats, and other species. Currently we have optimized protocols for up to nine-color staining (8 tyramides + DAPI), but the upper limit is likely higher given suitable dyes and imaging systems.
Rapid manual or automated protocol
Automation is also facilitated by the rapid protocol and unique features. The use of primary antibodies and our HRP quenching buffer makes it especially amenable for automated staining. External partners have developed a protocol on the Leica Biosystems BOND RX IHC staining system, and protocols for other systems are in development.

Pushing the boundaries further we have recently launched the SpectraSplit® 10 filter set, enabling imaging of up to ten colors without spectral unmixing. Again our design principle has been to make it an open solution, aligning the channels with most commonly used dyes from a variety of manufacturers.

Yes, StreptaClick® HRP heat-free TSA is easy to use and well-suited for those new to the field. The heat-free protocol considerably reduces the optimization time required to build a multiplex antibody panel.
Through its TSA and HRP block system, the need for secondary antibodies is eliminated. This improves reproducibility and streamlines the workflow compared to indirect immunofluorescence (IF), while preserving target epitopes and tissue morphology.
Up to 5-7 antibodies can be applied within a single day depending on the required incubation time of each antibody, significantly reducing the total experiment time. On average one staining cycle takes 1.5 hours.
250 reactions is the number reactions in singleplex that the kit is sufficient for. If all reactions were to be used for 5-plex experiments, the kit is sufficient for 250/5, or 50 five-plex reactions.

The reason more tyramide dyes are included in the 3 and 5 color kits is that some optimization may be required for the tyramide concentration; ex. some users prefer 1/50 dilutions, and sometimes 1/200 is sufficient.
Yes, StreptaClick® HRP heat-free TSA has been developed and validated for use with biotinylated primary antibodies. Biotinylated primary antibodies can be purchased from a number of vendors including our listed partners, either off the shelf or as custom orders. Alternatively, carrier free antibodies (ie no carrier protein such as BSA, and ideally free from sodium azide) can easily be biotinylated with our BiotinPure™ Antibody Biotinylation Kit. BiotinPure™ is an easy and complete solution only requiring a benchtop centrifuge.

StreptaClick® HRP Multiplex IHC kits
The kits contain StreptaClick® HRP antibody labeling reagent for labeling of biotinylated antibodies, a novel horseradish peroxidase (HRP) Quench Buffer (now also available as a standalone product), and Tyramide Amplification Buffer Plus (compatible with all common tyramide dyes from various vendors).
The staining protocol does not require heat treatment such as boiling between cycles, and is ideal for direct labeling of primary antibodies for higher degrees of multiplexing compared to working with secondary antibodies. Compatible with FFPE and frozen tissue sections, and fixed cells.
The number of reactions equals the number of possible singleplex TSA staining reactions when using the standard dilutions of the protocol. Divide by the number of markers in multiplex for the number of possible slides, ex 250/5 = 50 five-plex slides.
Tyramide Dye kits
All tyramide dye kits include PBS buffer and anhydrous DMSO to prepare tyramide stock solutions.
- Tyramide 8-Dye kit (800 reactions): includes 100 reactions each of Tyramide CF405S, Tyramide CF®430, Tyramide 488, Tyramide 555, Tyramide 594, Tyramide 647, Styramide iFluor 700 and Styramide iFluor 750 when used at the recommended 1/50 working dilution (subject to user optimization) for a total of 800 singleplex reactions.
